IGF-1 LR3 is a modified version of natural IGF-1 featuring two alterations: the substitution of glutamic acid (E) with arginine (R) at position 3 and the addition of a 13-amino acid extension at the N-terminus. These modifications significantly reduce binding affinity to IGF-binding proteins (IGFBPs), extending the half-life from approximately 10 minutes (for natural IGF-1) to 20–30 hours, while increasing in vitro biological activity to roughly 2–3 times that of natural IGF-1.
